During my research career I have been supported by generous WT and MRC Fellowships, and although clinically trained, my research was initially in basic science. Work on RNA processing and programmed cell death allowed me to develop molecular and cellular skills across a wide range of biological systems. I then decided, with WT support, to make a long-term commitment to the application of molecular and cellular immunology to dengue, a life-threatening global health issue. Dengue is endemic in > 100 countries, with SE Asia and the Western Pacific the most seriously affected. Explosive outbreaks can literally overwhelm healthcare systems; WHO estimate 20% of the world s population to be at risk, with around 50 million infections each year. It has taken a number of years to form collaborations to access clinical material and to develop our skill set in the many techniques needed to approach dengue on a broad front. We have considerable experience in the analysis of cellular and latter ly serological responses. These programmes are buttressed by our ability to work with the virus and although conceptually simple, it has taken several years to set in place a robust suite of virological assays. The laboratory is now uniquely poised to take the first steps in the early translation of these findings. The overarching aim of this proposal is to gain a better understanding of the pathology and immunology of dengue to inform the rational design of treatments and vaccines to combat d isease.
