The structural basis of the binding specificity of the family of SIRP paired receptors. (360G-Wellcome-084194_Z_07_Z)
The structural basis of the SIRP family of receptors will be determined. (i) X-ray crystallography will be carried out on the ligand binding domains of the SIRP beta and gamma (SIRP alpha has already been completed) (ii) The structure of the complex between CD47 and SIRPalpha will be determined working on extensive preliminary data. (iii) The structure of the three extracellular domains of SIRP alpha will be determined to work out the orientation of the interaction when cells interact; is i t compatible with cis interactions or close contacts with dimensions of the immunological synapse? (iv) Does the SIRP beta dimerise? Is there any evidence of association with between SIRP members? (v) Where is the mouse SIRP beta localised? (vi) Where are the more distantly related SIRP family members expressed and do they bind CD47?
