Development of a novel vaccine against Plasmodium vivax malaria using adenovirus and Modified Vaccinia Ankara (MVA) as recombinant vaccines. (360G-Wellcome-097395_Z_11_Z)

I will apply a leading vaccination strategy consisting on using clinically relevant adenoviral and MVA viral vectors to stimulate immune responses against Plasmodium vivax in order to prevent infection, disease and relapse through the elimination of pre-erythrocytic or liver-stage parasite forms. The key goals are: - Development of a pre-erythrocytic vaccine using adenovirus and MVA vectors expressing the circumsporozoite (CS) antigen and the thrombospondin-related adhesion protein (TRAP). - Assessing the immunogenicity of these vaccines. The latter will be done in collaboration with Dr. Socrates Herrera who is director of an NIH-funded International Center of Excellence for Malaria Research in Colombia and will permit assessing the protective efficacy in a P. vivax malaria sporozoite challenge in animal models using a system established by himself - Assessing the in vitro potential of these vaccines to eliminate the dormant hypnozoite forms. - Development of a novel rodent challenge system using mouse-specific transgenic P. berghei parasites expressing the P. vivax antigens PvCS and PvTRAP. - Development of a novel particulate antigen for a P. vivax vaccine, similar to that produced by GSK for P. falciparum malaria RTS,S and use of this in combination with viral vectors to enhance protection.
