The genetics of depression in diverse populations (360G-Wellcome-212360_Z_18_Z)
Major depressive disorder (MDD) is common and has an immense burden of disease. Genetic variants associated with MDD have been identified. However, most previous studies were based on European ancestry samples. Our aim is to establish a global study resource to investigate the genetic basis of depression in diverse ethnic groups. Due to the high prevalence of MDD, the rising numbers of biobanks and cohorts with health record linkage provide a unique and cost-effective opportunity to build a resource of diverse ancestry samples from existing studies. Our expected sample size includes > 26,500 affected and 764,000 unaffected individuals. We will use logistic mixed models to assess whether MDD loci identified in European samples have consistent effects across ethnicities. This question has profound implications for the transferability of findings. It will inform basic concepts of depression and could eventually affect clinical practice for diverse ethnic groups. The increased genetic diversity of our data resource can empower the discovery of novel genetic associations. Therefore, we will carry out trans-ethnic analysis to identify variants affecting MDD risk. These can point to biological pathways involved in the development of MDD and illuminate the aetiology of this prevalent yet still poorly understood disorder.
