Human Genetics and Disease Biology: Core Renewal for the Wellcome Trust Centre for Human Genetics 30 Oct 2016

Human genetics provides a unique and powerful tool for understanding normal biology, disease pathophysiology, and infectious mechanisms. The "genetic revolution" of the last decade has witnessed an explosion in knowledge of associated variants for individual traits. Looking forward, large population resources will facilitate a new paradigm, going from variants (individually or collectively) to multiple phenotypes, and new functional tools will help unlock causal genes and mechanisms. In parallel, genome sequencing will become routine in parts of clinical medicine. The Wellcome Trust Centre for Human Genetics (WTCHG), a large interdisciplinary research centre comprising 400 scientists in ~45 research groups, is one of the leading institutes, globally, in human genetics. Since its founding 21 years ago, the WTCHG has played a pioneering role in the progress and success of human disease genetics and mechanism research. Our focus is the development and implementation of novel approaches to exploit human genetics and uncover disease biology so as to improve healthcare. Critical to the Centre’s success is its lively environment which fosters collaborations between research groups across disciplinary boundaries, with research supported by seven Core teams. This application seeks core support to enable the WTCHG to continue its leading role in the field.

Wellcome Centre for Integrative Neuroimaging 30 Oct 2016

Understanding how interactions between neurons generate human behaviour, why individual brains vary from one another, or whether a patient is likely to develop a particular disease, requires explanations that span vast differences in scale. Yet such explanations are essential if insights from neuroscience are to make a meaningful impact on human health. Precise mechanisms discovered in animal models must be related to clinical phenotypes discovered through population studies; both must be combined to improve diagnosis and treatment in individual patients. Neuroimaging offers a powerful route to connect these different scales, providing measurements that are sensitive to cellular phenomena and that can be acquired in living humans. The WT Centre for Integrative Neuroimaging will enable novel insights into brain function that span levels of description, and therefore bridge the gap between laboratory neuroscience and human health. This will require fundamental discoveries concerning relationships between species and between scales, and major technological developments for mapping big-data discoveries onto neurobiological mechanisms. We will bring together diverse investigators who can tackle different themes within this grand challenge. Within each theme, neuroimaging will be used alongside complementary methodologies, ensuring that it takes inspiration from, and has impact on, areas beyond its typical reach.

Core Support for the East African Major Overseas Programme 30 Jun 2016

Our application provides the platform for a uniquely inter-disciplinary scientific programme linking biomedical, social and health systems research to deliver scientific insights of global importance to human health. We will work based from Kenya (Kilifi and Nairobi) and Eastern Uganda (Mbale). The Kilifi Programme will tighten its focus on our integrated platform (i.e. linked hospital/demographic/molecular surveillance) and legacy of continuous epidemiological data and stored samples over 25 years. Work in Mbale will consolidate a leading centre of clinical investigation in an area of hyper-endemic malaria transmission, and the Nairobi Programme will increase its independence with a focus on international disease mapping and health systems. Our major scientific themes include vaccines (including pre and post-licensing studies with exploratory immunology and epidemiological components), genomics and infectious disease transmission, clinical research (focusing on multi-centre clinical trials and the pathophysiology of critical illness with a developing programme on neonatal and maternal health), public health (with an emphasis on spatio-temporal analyses) and health systems research. The Programme is delivered by 29 PIs (i.e. scientists with independent funding). Training is central to our vision, and additional awards support 18 post-doctoral scientists and a projected 50 PhD students during the next 5 years.

Thailand Major Overseas Programme core grant renewal 2015-2020. 21 Apr 2015

The mission of the Wellcome Trust Thailand Major Overseas Programme is to carry out targetedclinical and public health investigations, using the best science, to provide appropriate andaffordable interventions which produce measurable improvements in the health of resource-poorpopulations in the tropics. Over the past 35 years the Programme has built substantial clinicaland laboratory research capabilities, deployed across a well-integrated network of strategicallyplaced permanent study sites and research units in seven countries across Asia and Africa. Wewill use this expertise and research capacity to tackle the epidemiology, diagnosis,pathophysiology, treatment, prevention, and, in the case of malaria, elimination of thoseunderstudied infectious and nutritional diseases that cause significant morbidity and mortality inthe populous rural tropics. In doing so we will strengthen local research capacity, a majorobjective of the programme, and build lasting south-south research collaborations with ourpartners across the developing world. This proposal is for core funding for an integrated programme of health research conducted in East Africa through the KEMRI Wellcome programme. Core funding supports the essential infrastructure and personnel to provide a platform on which the specific research components, are built. The research programme it self is embedded in a national research institute (KEMRI) but has collaborative links to a large number of governmental, educational and research institutions in Kenya and the East A frican region . The research programme is conducted by 32 principal investigators, funded by competitive research grants and fellowships, working closely together under four main disciplines: (1) Clinical Sciences (2) Epidemiology, Demography and Public Health (3) Pathogen Biology (4) Health Systems and Social Science Research. For the period 2011-2016 the programme has four key strategic scientific themes: Improving child survival during a period of rapid epidemiological transition Ad dressing adult health focussing on maternal health and recognising the impact of HIV on vulnerable populations Strengthening health systems through research on policy for, access to, monitoring, evaluation and delivery of care. Understanding the genetic basis of common human diseases: core funding for the Wellcome Trust Centre for Human Genetics. 22 Feb 2010

The Wellcome Trust Centre for Human Genetics was established in 1994 to undertake research into the genetic basis of common diseases. The objective of the Centre is to gain insight into mechanisms controlling genetic susceptibility to human disease, including the localization and identification of disease genes or disease-causing variants, functional characterization of genetic variants responsible for susceptibility, understanding how they contribute to disease risk in populations and how genet ic factors contribute biologically to disease processes, and the development and application of new analytical tools. In order to achieve this objective the Centre has brought together multidisciplinary research groups collaborating on human and rodent genetics, genetic epidemiology and statistical genetics, functional analysis of disease genes, and structural biology. The Centre has a strong focus of expertise, equipment, and resources centralized in Core groups allowing all research groups to benefit and expand their research strategies. Recent years have seen major advances in human genetics, in which Centre scientists have played leading roles, and we see major opportunities and challenges ahead. This application is for the renewal of the 5-year core award to fund the Centre's Core Groups: Genomics, Bioinformatics, Molecular Cytogenetics and Microscopy, Transgenics, and IT, for the period 1.4.2011 to 31.3.2016.

Viet Nam Major Overseas Programme Core Grant Renewal 2010-2015. 21 Sep 2009

Infectious diseases remain a very real global threat. In addition to the endemic infections there has been emergence of new, often zoonotic, diseases and the rise of drug resistance among all major pathogens. Asia is the epicentre of these changes. Within a four hour flight of Vietnam live half the world s human population and 75% of the world s poultry. The region is a global hot spot for the emergence of new infectious diseases, it leads the world in drug resistance, and is going through an unprecedented period of social and environmental change. The future impact of these changes is difficult to predict but it will influence disease patterns both directly and indirectly. We believe that this trusted collaboration between Vietnam and the Wellcome Trust is uniquely placed to address these complex issues both nationally and regionally. Our work keeps health research as the focus with an integrated science programme and a particular interest in the interface between human and animal health. Our training and public engagement programme from schools to post-docs and lay people is helping to build a critical mass of Asian clinicians and scientists dedicated to the development of the region and the health of its people.

Close integration with host institutions, a broad capability, and the capacity to respond rapidly and effectively to emerging threats, has placed the unit in the forefront of research on rural febrile illnesses, the main cause of hospital admission in tropical countries, and drug-resistant malaria, the main threat to global malaria control and elimination. We have created a network of like-minded research collaborations across Asia and Africa, and we foster an international c adre of physicians and scientists with a common goal of providing immediate and measurable practical solutions to the major health problems confronting the developing world. We believe this multinational, integrated, adaptive, and evolving model based on sound science and clinical excellence is the most effective means of delivering research to improve health.
